Inhibition of immunoglobulin E attenuates pulmonary hypertension

چکیده
Pulmonary hypertension (PH) is a severe cardiopulmonary disease characterized by pulmonary vascular remodeling. Immunoglobulin E (IgE) known to participate in aortic remodeling, but whether IgE mediates unknown. In the present study, we found serum elevation arterial (PAH) patients, hypoxia-induced PH mice and monocrotaline-induced rats. Neutralizing with an anti-IgE antibody was effective preventing development rat models. The receptor Fc?RI? also upregulated lung tissues Fcer1a deficiency prevented of PH. Single-cell RNA-sequencing revealed that mostly expressed mast cells (MCs) MC-specific knockout protected against mice. IgE-activated MCs produced interleukin (IL)-6 IL-13, which subsequently promoted muscularization. Clinically approved omalizumab alleviated progression established Using genetic pharmacological approaches, have demonstrated blocking IgE–Fc?RI? signaling may hold potential for PAH treatment. mouse models patients’ data, Shu, Liu, Zhou et al. show concomitant increase immunoglobulin expressing effector has important role inhibition animal

Inhibition of phosphodiesterase-1 attenuates cold-induced pulmonary hypertension.
Chronic exposure to cold caused pulmonary arterial hypertension (cold-induced pulmonary hypertension [CIPH]) and increased phosphodiesterase-1C (PDE-1C) expression in pulmonary arteries (PAs) in rats. The purpose of this study is to investigate a hypothesis that inhibition of PDE-1 would decrease inflammatory infiltrates and superoxide production leading to attenuation of CIPH. Three groups of ...
متن کاملPDE5A inhibition attenuates bleomycin-induced pulmonary fibrosis and pulmonary hypertension through inhibition of ROS generation and RhoA/Rho kinase activation.
Pulmonary hypertension frequently complicates interstitial lung disease, where it is associated with a high mortality. Patients with this dual diagnosis often fare worse than those with pulmonary arterial hypertension (PAH) alone and respond poorly to standard PAH therapy, often dying of right ventricular (RV) failure. We hypothesize that nitric oxide synthase (NOS) uncoupling is important in t...
متن کاملMitochondrial transplantation attenuates hypoxic pulmonary hypertension
Mitochondria are essential for the onset of hypoxia-induced pulmonary vasoconstriction and pulmonary vascular-remodeling, two major aspects underlying the development of pulmonary hypertension, an incurable disease. However, hypoxia induces relaxation of systemic arteries such as femoral arteries and mitochondrial heterogeneity controls the distinct responses of pulmonary versus femoral artery ...
متن کاملMineralocorticoid receptor antagonism attenuates experimental pulmonary hypertension.
Mineralocorticoid receptor (MR) activation stimulates systemic vascular and left ventricular remodeling. We hypothesized that MR contributes to pulmonary vascular and right ventricular (RV) remodeling of pulmonary hypertension (PH). We evaluated the efficacy of MR antagonism by spironolactone in two experimental PH models; mouse chronic hypoxia-induced PH (prevention model) and rat monocrotalin...
متن کاملEvaluation of Pulmonary Hypertension In First Degree Relatives Of Patients With Primary Pulmonary Hypertension
Background: Pulmonary hypertension (PH) was defined for the first time in 1951 as primary pulmonary hypertension (PPH). Some studies emphasized on the role of genetics in the development of pulmonary hypertension in family members of affected patients. So, in this study we evaluated the prevalence of pulmonary hypertension in first degree family of patients with documented PPH. Methods: In thi...
